The biggest mistake that most first-time job applicants make is forgetting to interview the company that's interviewing them. Don't just ask the generic "So what's it like to work here?" question. You won't get the clear picture you need. Ask for anecdotes and stories that describe what makes the company culture unique: "What happens here that wouldn't happen elsewhere?" That way, you'll get a better understanding of what the day-to-day workplace might be like. After all, it's just as important that the company be a good fit for you as the other way around.
